To identify genes that might be relevant to the cytotoxic effect of the anticancer drug 3-bromopyruvate (3BP), which is cytotoxic to prostate cancer cells through the induction of apoptosis, we analyzed a cDNA array of LNCaP cells. Nineteen genes out of the 2718 cDNA clones of the array exhibited expression changes following exposure to 3BP. We verified the expression of 3 genes (CEACAM3, CLU, DCXR) by Northern blot. None of the genes were related to cell cycle. We found increased expression of some genes (IL-8, GADD153/CHOP) in 3BP-treated cells, and decreased expression of other genes (CEACAM3, FADD/MORT1) in 3BP-treated cells. The expression of CEACAM3 increased in TNF-alpha-treated cells and in cells that were not exposed to 3BP, but this was not accompanied by caspase activation. We suggest that CEACAM3 is not an early marker of apoptosis, and that the downregulation of this gene is not relevant to 3BP-induced apoptosis.The present disclosure generally relates to protein-based compositions and methods for preparing and using the same.
